Physical and Chemical Properties | Bleomycin contains several components with similar properties and is a weakly alkaline substance. Soluble in water, methanol, slightly soluble in ethanol, almost insoluble in acetone, ethyl acetate, butyl acetate, ether. It is blue-green when containing copper ions, and white powder after removing copper ions. |
Use | The recommended concentration as a selector is 10-100 μg/ml. Bbleomycin is used as a selector for transformed cells, especially for the transformation of plant cells. |
In vitro study | Both UT-SCC-12A and UT-SCC-12B were resistant to Bleomycin sulfate with IC50 of 14.2 nM and 13 nM, respectively. Alveolar macrophages cultured with 0.01 μg/mL to 1 μg/mL Bleomycin sulfate for 18 hours secreted significantly more fibroblast growth factor than macrophages cultured alone. Macrophages stimulated by Bleomycin sulfate continued to produce large amounts of fibroblast growth factor, even after Bleomycin sulfate was removed and replaced with fresh medium (without Bleomycin sulfate). Stimulation of alveolar macrophages by Bleomycin sulfate leads to inhibition of cellulose growth factor secretion by cycloheximide, and the 5-lipoxygenase inhibitors NDGA (nordihydroguaiaretic acid) and BW755c, it is shown that it requires not only protein synthesis but also the complete expression of the activity of the 5-lipoxygenase pathway of arachidonic acid metabolism. Bleomycin sulfate (400 µg/mL), cultured for 24 hours, decreased the activity of NTera-2 cells, increased the activity of caspase-3,-8 and -9, increased the levels of Bax and cytoplasmic cytochrome C, and reduce Bcl-2 levels. For instability aberrations, the chromosomal disrupting effect of Bleomycin sulfate on ADIPO-P2 cells persisted for at least 10 days after treatment. Furthermore, the appearance of telomere fusions after 10 days of treatment of cells exposed to Bleomycin sulfate suggests that Bleomycin sulfate is capable of inducing delayed telomere destabilization. |
